Nata in Scozia, la giovane Annie frequentò una scuola per allievi particolarmente intelligenti. Ha studiato anche il flauto traverso alla Royal Academy of Music di Londra. Ha raggiunto la popolarità dopo aver cantato per tre anni con i The Tourists (con cui ha inciso 3 LP). Negli anni 1980 tra Annie e il chitarrista del gruppo, Dave A. Stewart, nasce una relazione sentimentale e i due decidono di lasciare il gruppo per crearne uno nuovo: gli Eurythmics. All'inizio nella carriera degli Eurythmics, la Lennox era conosciuta per il suo modo di vestire androgino, che una volta impersonò anche Elvis Presley. Molti giornalisti spesso si riferivano a lei come "la Grace Jones bianca". Questo duo negli anni 80 ha pubblicato una lunga serie di singoli classici e molto famosi, come: "Sweet Dreams (Are Made Of This)", "Here Comes The Rain Again", "Would I Lie To You?", "There Must Be An Angel (Playing With My Heart)", "Missionary Man", "You Have Placed A Chill In My Heart", "Don't Ask Me Why" e altri. Nel 1991, dopo la rottura con Dave Stewart, ha intrapreso una lunga carriera da solista con eguale successo. Il suo primo album solista del 1992, Diva, fu un successo inequivocabile sia di critica che commerciale: il suo successo continuò grazie ai singoli estratti da Diva, a numerosi premi e ad una colonna sonora per il film Dracula di Bram Stoker di Francis Ford Coppola del 1992. Nello stesso anno partecipò anche al Freddie Mercury Tribute Concert duettando con David Bowie nella canzone Under Pressure. Le sue apparizioni si fanno però sempre più rare e questo contribuisce a farla uscire progressivamente dallo show business. Medusa venne pubblicato tre anni dopo Diva ed ha ottenuto comunque buoni risultati di vendita. Nel 1997 Annie ha registrato nuovamente la canzone degli Eurythmics Angel per in omaggio a Diana, Principessa del Galles. Nel 1998 ha stabilito nuovamente i contatti con Dave Stewart e nel 1999 i rinati Eurythmics hanno prodotto l'album Peace. Nel 2003 ha pubblicato il suo terzo album da solista: Bare. Nel 2004, la Lennox ha vinto l'Oscar per la migliore canzone (Academy Award for Best Song) per Into The West dal film Il Signore degli Anelli: Il Ritorno del Re alla 76th Academy Awards. Precedentemente aveva registrato Use Well the Days per il film, nel cui testo abbondano le citazioni di Tolkien. Questa canzone non è stata utilizzata nel film, bensì appare su un DVD bonus incluso con l'edizione speciale del CD della colonna sonora del film. Il suo nuovo album si chiama Songs of mass destruction ed è uscito in Italia il 1 ottobre 2007. Nel luglio 2005, Annie Lennox ha partecipato al Live 8 ad Hyde Park, Londra. Annie Lennox è stata elogiata dai critici di musica e di video femministi e gay. Nel 1991 ha partecipato con un cameo al film a tematica LGBT Edoardo II di Derek Jarman. Pare che durante gli anni d'oro degli Eurythmics, le ragazze più giovani la ammirassero più della regina di Inghilterra. Interprete raffinata ma anche potente, soffre di affaticamento alle corde vocali e questo le impedisce di fare frequenti concerti dal vivo. Annie, inoltre, è notoriamente difficile da intervistare ed ha sofferto di depressione maniacale. Ha due figlie ed ha divorziato due volte.
